Serum Lactic Dehydrogenase Activity: an Analytical Assessment of Current Assays.
نویسندگان
چکیده
Serum LDH activities assayed spectrophotometrically by the “forward” (lactate + DPN to pyruvate + DPNH + H) method have linear reaction rates, activity proportional to the amount of serumadded, anda reproducibilityof 1.3%. The reagent mixtore can be stored frozen for 6 monthsand in the lyophilized state indefinitely. Poor separationof normal from abnormal serum LDH activities results from the nonlinearcalibrationcurves anddecreasingreaction rates of the “backward” spectrophotometric assay. The dinitrophenylhydrazinecolorimetric assay is deemed unsuitable for clinical use because largevariationsin activity result from minor variations in the reagent calibration curve, and because colorimetric activities are inaccurate as compared to activities obtained with the “forward” spectrophotometricassay.

Correlation of selected serum constituents: 1. Inter-individual variation and analytical error.
The intra-subject correlations of three clinically meaningful combinations of serum constituents--(a) potassium, calcium, and albumin; (b) urea, creatinine, and uric acid; and (c) aspartate aminotransferase, lactate dehydrogenase, and alkaline phosphatase--were studied in 11 healthy men. Duplicate serum samples were obtained at 800 h, 1100 h, and 1400 h on five different days. All assays were p...

متن کاملA response to "Serum-ethanol determination: comparison of lactate and lactate dehydrogenase interference in three enzymatic assays".
Gas chromatography is considered to be the reference method for ethyl alcohol determination. However, enzymatic ethanol assays have been developed for use in the clinical laboratory by several commercial vendors. Essentially, these assays utilize the oxidation of ethyl alcohol to acetaldehyde with concurrent reduction of nicotinamide adenine dinucleotide (NAD) to NADH while monitoring the incre...
متن کاملAn automated colorimetric (tetrazolium salt) assay for serum lactate dehydrogenase.
The adaptation of a tetrazolium salt assay for serum lactate dehydrogenase to full automation is described. The tetrazolium salt, INT, (2-p-iodophenyl-3-p-nitrophenyl5-phenyl tetrazolium chloride) serves as the chromogenic indicator of the enzyme activity. Phenazine methosulfate serves as an intermediate electron carrier. Estimation requires only 5 mm. of incubation, and assays may be run at th...
